For the uninformed, this is the best discussion on the subject of DNA that I have ever seen. I have been trying to determine who my great great grandfather is for years. I’ve tested with Ancestry and Family Tree DNA, hired ProGenealogists with Ancestry (twice), and still can’t determine who he is. I truly don’t know where to go now. The genealogist that consults with Finding your Roots works for a company that doesn’t do individual research. Who else does the genetic genealogist research that they do?
I hope this helps to clear things up. Ancestry DNA testing is not an exact science, and is limited by the fact that we don't inherit the exact same DNA our parents had, meaning that with each new generation, old DNA is lost. Ancestry tests can provide estimations of our genetic ancestry, and though they are improving all the time, they can't tell the whole story of our heritage.
While DNA contains material common to all humans, some portions are unique to each individual. These portions, or regions, contain two genetic types (alleles) that are inherited from the person’s mother and father. A person’s DNA profile is made by investigating a number of these regions. In a paternity test, for example, the mother’s DNA profile is compared with the child’s to find which half was passed on by the mother. The other half of the child’s DNA is then compared with the alleged father’s DNA profile. If they don’t match, the ‘father’ is excluded, which means he isn’t the father of that child. If the DNA profiles match, the ‘father’ is not excluded - which means there is a high probability (more than 99 per cent) that he is the father. DNA tests such as this can’t offer 100 per cent proof.
Many people who submit their DNA to a direct-to-consumer company also upload their raw information to public databases like GEDmatch, which law enforcement can access. People upload their raw DNA data after taking another test, like those from 23andMe or Ancestry, to several open online DNA databases. Most companies do not release database information to law enforcement. However, a recent study found that, using publicly available data, it's possible to identify up to 60 percent of Americans with European heritage via third-cousin-or-closer DNA.
Specific tests for your father’s family include ‘Y-DNA’ tests which focus on the ‘Y chromosomes’ in your cells’ nuclei, passed down from father to son. Specific tests for your mother’s family include ‘mtDNA’ tests which report on a subset of DNA found in the ‘mitochondria’ (your cells’ energy factories), passed down from mother to son or to daughter.
When I found out about AncestryDNA, I thought this could be the perfect tool to pinpoint where my family emigrated over the past few hundred years (AncestryDNA can actually go back 1000 years) and give me a focus where to take my search next. Three of the companies, MyHeritage, Ancestry and FTDNA, use the Illumina OmniExpress chip and 23andMe uses the new Infinium® Global Screening Array chip from Illumina. The fact that all of the chips come from the same company may be confusing, leading some to believe that all tests are created equal. This is not the case. The chip used to process DNA samples is only one part of the process. Each company develops their own analysis of the results, references different population samples and provides different reports. In addition, each one of these DNA test providers offers different tools for you to analyze the data you receive, creating variations in results, accessibility and usefulness.

Health and disease info: DNA testing can also indicate which conditions for which you may have a preponderance. It's a controversial feature, to be sure. Knowing that you have a genetic predisposition to a certain form of cancer may make you more vigilant for testing, but it may also lead to increased stress -- worrying about a potential condition that may never develop, even if you're "genetically susceptible" to it. The possibility of false positives and false negatives abound -- any such information should be discussed with your doctor before you act upon it.
For better ancestry and medical insights, you should encourage family members, especially parents and grandparents, to take a DNA test as well. If your family is from a specific geographical location for generations, your samples could potentially improve the service's reference panel, in turn improving results for everyone. If you’re female and take a test from 23andMe or LivingDNA, you can view paternal haplogroup information, and you get more information when one of your male family members takes a test as well.
I once heard someone explain DNA as being like a cake recipe book in a library. You can take the book out of the library and even unravel it by taking out the pages. You follow the cake recipes inside the book but when ever you make the cake it never turns out quite the same way twice. When you've finished you have to return the recipe book to the library because that is where its stored. Made sense to me at the time
Because it is a genetic condition, FH is present at birth, meaning most people with this condition have high LDL cholesterol levels from a young age. There are many places you can upload your raw DNA, and several of them are free. Popular third-party DNA analysis tools include GEDmatch and Promethese. GEDmatch is a free, open database and genealogy site that gives additional DNA relative matching and trait results. This tool has information from users of multiple different testing companies. Promethease compares your raw DNA information against scientific reports that link certain markers to health conditions, though you should take these results with a grain of salt as genetic links do not equal a diagnosis.
4. Geno 2.0: At $199 National Geographic’s Genographic Project is the most expensive of all of the choices and is also not deigned specifically for genealogists. They do not offer a database of known relations and the data they provide is more general in nature. However, joining the project does provide a geographic ancestry breakdown as well as a report on how much Neanderthal you may have (fun!). It is an exciting scientific endeavor and definitely worth exploring and joining if you have the resources to do so. Find it here.
