Most of the services we tested use genotyping to read your DNA. Genotyping looks for specific markers in your genetic code. For something like ancestry testing, genotyping is effective because it identifies known variants in your DNA. Scientifically speaking, genotyping’s weakness is that it can only recognize previously identified markers. This is one reason DNA tests’ accuracy relies so heavily on the DNA database size; there must be enough information available and identified genetic variants in the database to recognize new customers’ markers.
At-home paternity tests have been around much longer than other direct-to-consumer DNA tests. Most of them require you to collect cheek swab samples from a prospective father and child, which you then send off to a lab to determine paternity. For non-legal use, these tests can cost as little as $15, but tests that provide verified results that are admissible in court cost a few hundred dollars. 
Although, I have absolutely no British or Irish ancestry, I found my results extremely satisfying. I particularly appreciate that living DNA gives you a lot of ways to view your data. You can see your ancestry results as color-coded dots filling up a person’s silhouette, on a map, as a pie chart or on a timeline. All the graphics present the same set of data, but each has its own appeal. Within each graphic, you can also choose to view global or regional matches and cautious, standard or complete estimates, which each have a different level of detail and certainty.

The reason that saliva works as well as blood (or hair follicles or skin samples) is that your DNA -- which is short for deoxyribonucleic acid -- is present in all of them. It's the basic genetic code present in all of your cells that makes up your key attributes, from the color of your eyes to the shape of your ears to how susceptible you are to cholesterol.
I’ve had the same experience, and so have many others. My mother’s family is all from Italy, and yet my results came back with NO Italian whatsoever. Another said there was. None of them report German as a result, which is quite strange since Germans are definitely a people! These DNA tests are subjective and based on human analysis. As we all know, humans make mistakes. At the end of it all, I’ve decided that I’ll just stick with the ancestry my grandparents told me about when they were alive.
Because it is a genetic condition, FH is present at birth, meaning most people with this condition have high LDL cholesterol levels from a young age. Since many people with FH show no physical symptoms, this condition is typically diagnosed with a blood test for cholesterol. However, some people with FH may not be diagnosed until after experiencing symptoms related to early heart disease, including chest pain or heart attack.
As a postscript, I eventually end up having an interesting chat with Titanovo about my “bioinformatics” (distilled from my 23andMe data). One of the first things I’m told is that my eyes are green (they’re brown). However, the bioinformatics got my skin type and frame/weight generally right and had interesting (albeit occasionally generic) things to say about exercise, diet, goals, steering clear of too much sugar and so on.
Whole genome sequencing is the most accurate representation of your DNA that you can buy, as it provides you with the details of every single base in your DNA (more than 5 billion). This is unsurprisingly much more expensive than DNA profiling, and isn’t necessary if you are looking for a profile for identification purposes. If you are interested in doing in-depth analysis, the firm offers a chromosome browser, allows raw data to be uploaded, provides support for setting different segment matching thresholds, and allows up to five comparisons to be done at once. Family Tree DNA allows trial transfers from 23andMe and AncestryDNA into its match database; additional transfers of various datasets is available for a fee. The company promises to keep data for 25 years.
Next, you'll receive an email alert that your results are ready, and that's when the fun begins. Your results may not be as dramatic as those portrayed in TV ads, but you may find some surprises. One important note: Results are different for women and men. Women, who have the XX chromosome, can only trace back the maternal line. Men, having the XY chromosome, can track back the maternal and paternal line, painting a complete picture. If you're a woman, it's worth asking your brother, if you have one, to take a test and share the results. When some of these services ask for your sex when you order your kit, they simply want to know about your chromosomes.
Nacho Esteban of 24Genetics told us, “Ancestry is not an exact science. The top five companies in the world would show very similar results when talking about continents; the similarity is smaller when talking about countries. In regional ancestry, some border regions are difficult to identify and sometimes there may be discrepancies. So we cannot take the information as something 100% sure. But at the end, it gives a great picture of where our ancestors were from.”

The case of Colin Pitchfork, the first criminal convicted using DNA fingerprinting, is well publicised, but the very first use of this technique was actually in an immigration case. Alec Jeffreys (the person who developed the technique) helped a Ghanaian boy to avoid deportation by comparing his DNA to that of his alleged British mother’s, to prove that he was her biological son. Since that case, DNA profiling has been used in thousands of cases of immigration, by either keeping families together or reuniting them by proving biological relationships.

the beauty of a y-DNA test is that it tracks the paternal y-chromosome…..yes, even indicating a surname change but not when the surname changed [does not match known male descendants]. In all DNA testing, it really helps to have researched about 5 generations back on all lineages……that way you can find common surnames in the autosomal tests. The y-DNA tests go back for centuries…..and the autosomal testing really only goes back about 5 generations…….

Which is all very well, but do these kits work and deliver the service they promise and what about the wider ethics and implications of home genetic testing? Is it always wise for generally under-informed, under-prepared consumers to meddle in the highly complex, nuanced arena of genetics, risking confusion, complacency or even outright panic and anxiety when confronted with ostensible “bad news” (which may not even be true)?
After taking a DNA test for ethnicity, many ancestry companies will give you the option to contact their other customers, providing you share sections of your autosomal DNA with them (meaning that you’re related to these individuals to a greater or lesser degree). The companies that do this maintain ‘Family Finder’ databases, and the people you’re related to are usually referred to as ‘matches’. The three largest companies that do this are Family Tree DNA, Ancestry.com and 23andMe, and as part of the results that they provide, you’ll be able to see a list of these living relatives ranked according to how closely you’re related to them.
